Investigation of PRL-RsaI and HaeIII gene polymorphisms in Anatolian water buffaloes bred by using PCR-RFLP method R. Bras. Zootec.
Özşensoy,Yusuf.
ABSTRACT The objective of this study was to investigate polymorphisms both in exons 1 and 3 of prolactin (PRL) gene for milk productivity of Anatolian water buffalo breed in Sivas province in Turkey. Blood samples were collected from 129 male and female water buffaloes and DNA was isolated by using phenol/chloroform method. Samples of DNA were amplified and resulting PCR products were digested with RsaI (for exons 1 and 3) and HaeIII (for exon 1). Allelic polymorphisms were determined by separation of fragments obtained from digested PCR products in 3% agarose gel electrophoresis. AA genotype (HaeIII) and BB genotype (RsaI) of exon 1 and only AA genotype (RsaI) of exon 3 were obtained. No polymorphisms were determined in Anatolian water buffalo breed and...

Kappa-casein gene study with molecular markers in female buffaloes (Bubalus bubalis) Genet. Mol. Biol.
Otaviano,Antonio Roberto; Tonhati,Humberto; Sena,Janete Aparecida Desidério; Cerón Muñoz,Mario Fernando.
Caseins comprise make up about 80% of the total protein content of milk and present polymorphism with changes in the amino acid sequence. Within this abundance of proteins, kappa-casein is noteworthy, since it has been associated with differences in milk yield, composition and processing. The objective of this study was to observe the existence of polymorphism in the kappa-casein gene in female buffaloes. For this purpose, blood samples from 115 female buffaloes, collected with vacutainer by needle punctionure of the jugular vein, were used. for genomic DNA extraction was done from blood samples. The PCR-RFLP and SSCP techniques demonstrated that the studied animals were monomorphic for the kappa-casein gene. Only allele B was observed in these animals,...

Molecular assessment of Gymnotus spp. (Gymnotiformes: Gymnotidae) fishing used as live baitfish in the Tietê River, Brazil Neotropical Ichthyology
Faria-Pereira,Lilian Paula; Hilsdorf,Alexandre Wagner Silva; Albert,James; Paiva,Maria José Tavares Ranzani; Galvão,Márcia Santos Nunes.
ABSTRACT The capture of live bait for sport fishing is an important activity for fishing communities. The main species used for this purpose are members of the genus Gymnotus, which comprises numerous species of cryptic nature that are difficult to identify based on external morphology. The aims of this work were to identify through partial sequences of the COI gene Gymnotus species fished in the Jacaré-Guaçu River, SP, and to develop a molecular diagnostic approach using PCR-RFLP to identify these species. Partial COI sequences were compared to those of other species deposited in GenBank. The sequences were assessed in the NEBCutter program to determine restriction sites in the sequence and the enzymes to be tested. Phenetic analysis performed by...

Molecular Authentication of Meats from Three Terrestrial Birds Based on Pcr-Rflp Analysis of the Mitochondrial 12S rRNA Gene Rev. Bras. Ciênc. Avic.
He,H; Wang,Y; Qing,Y; Li,D; Zhao,X; Zhu,Q; Yin,H.
ABSTRACT In this study, a method utilizing PCR-restriction fragment length polymorphism (PCR-RFLP) of a mitochondrial gene was developed for the identification of chicken (Gallus gallus), quail (Coturnix coturnix), and common pigeon (Columba livia) meat. PCR products of ~440 bp were obtained from the 12S rRNA gene of these three birds using a pair of universal primers. The three terrestrial birds can be distinguished using one restriction endonuclease, Alu I, which was selected based on species-specific variations in the mt 12S rRNA gene sequence using 9 newly-obtained and 44 published chicken, quail and pigeon sequences. This method was also successfully used to identify commercial quail and pigeon meat products, which were found to be adulterated with...

Molecular characterization of enteroinvasive Escherichia coli ipa genes by PCR-RFLP analysis BJM
Gibotti,Adriana; Tanaka,Tânia L.; Oliveira,Valéria R.; Taddei,Carla R.; Martinez,Marina B..
In this study, polymorphism in ipa genes was found in five out of nine EIEC serotypes studied. When SalI and HindII were used in RFLP-PCR assays many EIEC serotypes showed polymorphism in ipaB and ipaD. On the other hand, no polymorphism was observed in ipaA and ipaC in these strains. The polymorphism present in EIEC strains is serotype-dependent, since restriction patterns were conserved amongst strains belonging to the same serotype. When IpaB deduced amino acid sequences of S. flexneri M90T and FBC124-13 were compared, ten amino acids changes could be observed mainly in the amino-terminal region. The deduced EIEC IpaD amino-acid sequence presented 91% similarity with the Shigella strain. In this case, amino acid changes were spread out through the whole...

Molecular identification of species of the Genus Euglossa Latreille (Hymenoptera: Apidae, Euglossini) Neotropical Entomology
López-Uribe,Margarita M.; Del Lama,Marco A..
Euglossine bees interact with more than 60 plant families of the Neotropical region. The richness and abundance of these bees have been intensively studied in different ecosystems using the methodology of capturing males with chemical baits. Females are poorly known for most of the species and morphological characters for their taxonomic classification have not yet been described. The purpose of this study was to use allozymes and restriction patterns of the mitochondrial regions 16S and Cyt b to identify species of Euglossa Latreille. Bees were collected while visiting Thevetia peruviana (Apocynaceae) flowers in five cities of the state of São Paulo, Brazil. Three Euglossa species were identified among the 305 individuals collected. Euglossa cordata (L.)...

Almeida,Álvaro M. R.; Piuga,Fernanda F.; Marin,Silvana R. R.; Binneck,Eliseu; Sartori,Fábio; Costamilan,Leila M.; Teixeira,Maria R. O.; Lopes,Marcelo.
Cercospora kikuchii, involved with the defoliation of soybean (Glycine max) plants, is normally associated with Septoria glycines in late season. Seventy-two isolates from different regions in Brazil, obtained mainly from purple stained seeds, showed phenotypic variation. Cercosporin content and rate of colony growth was higly variable among isolates. A strong correlation between cercosporin content and virulence was identified. Genetic variation among and within populations was evaluated based on 86 RAPD loci. The RAPD analysis clustered all isolates into seven groups. No relationship was observed between RAPD groups and geographic origin or cercosporin content. The sequence of the internal spacer regions (ITS1-5.8S-ITS2) from 13 isolates chosen according...

PCR-RFLP analyses of formalin-fixed fish eggs for the mapping of spawning areas in the Eastern Channel and Southern North Sea ArchiMer
Lelievre, Stephanie; Verrez-bagnis, Veronique; Jerome, Marc; Vaz, Sandrine.
A better knowledge on spawning grounds of principal commercial fishes from North Sea is necessary for their monitoring. Fish egg taxonomic identification based on morphological characters cannot always be carried out effectively. This is particularly the case for a number of Gadidae fish species eggs (i.e. cod, haddock and whiting, and three rockling species) which have the same range of size and for which molecular-based techniques can help improving the accuracy of egg taxonomic identification, and hence the reliability of studies on spawning areas. In this study, formalin fixation, egg conservation and DNA extraction were optimized in order to develop a specific PCR-RFLP method. Based on the sequence of the cytochrome b gene of reference specimens, a...

Polymorphism in the internal transcribed spacer (ITS) of the ribosomal DNA of 26 isolates of ectomycorrhizal fungi Genet. Mol. Biol.
Gomes,Eliane A.; Kasuya,Maria Catarina M.; Barros,Everaldo G. de; Borges,Arnaldo C.; Araújo,Elza F..
Inter- and intraspecific variation among 26 isolates of ectomycorrhizal fungi belonging to 8 genera and 19 species were evaluated by analysis of the internal transcribed sequence (ITS) of the rDNA region using restriction fragment length polymorphism (RFLP). The ITS region was first amplified by polymerase chain reaction (PCR) with specific primers and then cleaved with different restriction enzymes. Amplification products, which ranged between 560 and 750 base pairs (bp), were obtained for all the isolates analyzed. The degree of polymorphism observed did not allow proper identification of most of the isolates. Cleavage of amplified fragments with the restriction enzymes Alu I, Hae III, Hinf I, and Hpa II revealed extensive polymorphism. All eight genera...

Polymorphisms of the bovine chemokine receptor-like 1 gene and their associations with meat quality traits in Qinchuan cattle (Bos taurus) Electron. J. Biotechnol.
Zhan,Xiaoli; Jiang,Bijie; Ma,Xianghui; Fu,Changzhen; Zan,Linsen.
Background: We investigated the polymorphisms of the bovine chemokine receptor-like 1(CMKLR1) gene. The coding region of CMKLR1 was screened in Qinchuan cattle by PCR-RFLP technology. Results: In this study, we discovered two single nucleotide polymorphisms (SNPs) (264G > C and 762C > T) in the coding region of the CMKLR1 gene. Hence, we described the BmgT120l and Pdm1 PCR-RFLP methods for detecting the 64G > C and 762C > T mutations, respectively. PCR-RFLP and sequencing were used to analyze the two loci of CMKLR1 gene in 324 individuals, which were randomly selected from breeding populations. Furthermore, meat quality traits in another 80 Qinchuan individuals were analyzed by the comparison between the genotypes and their phenotypic data....

Rapid detection of Candida species in bronchoalveolar lavage fluid from patients with pulmonary symptoms BJM
Zarrinfar,Hossein; Kaboli,Saeed; Dolatabadi,Somayeh; Mohammadi,Rasoul.
Abstract Candida species, especially C. albicans, are commensals on human mucosal surfaces, but are increasingly becoming one of the important invasive pathogens as seen by a rise in its prevalence in immunocompromised patients and in antibiotic consumption. Thus, an accurate identification of Candida species in patients with pulmonary symptoms can provide important information for effective treatment. A total of 75 clinical isolates of Candida species were obtained from the bronchoalveolar lavage fluid of both immunocompromised and immunocompetent patients with pulmonary symptoms. Candida cultures were identified based on nuclear ribosomal Internal Transcribed Spacer (ITS1-ITS2 rDNA) sequence analysis by polymerase chain reaction–restriction fragment...

Reconstruction of major maternal and paternal lineages of the Cape Muslim population Genet. Mol. Biol.
Isaacs,Shafieka; Geduld-Ullah,Tasneem; Benjeddou,Mongi.
The earliest Cape Muslims were brought to the Cape (Cape Town -South Africa) from Africa and Asia from 1652 to 1834. They were part of an involuntary migration of slaves, political prisoners and convicts, and they contributed to the ethnic diversity of the present Cape Muslim population of South Africa. The history of the Cape Muslims has been well documented and researched however no in-depth genetic studies have been undertaken. The aim of the present study was to determine the respective African, Asian and European contributions to the mtDNA (maternal) and Y-chromosomal (paternal) gene pool of the Cape Muslim population, by analyzing DNA samples of 100 unrelated Muslim males born in the Cape Metropolitan area. A panel of six mtDNA and eight Y-chromosome...
